Moeller Decarboxylase Broth with Lysine is a differential biochemical medium used to determine whether microorganisms produce the enzyme lysine decarboxylase. It is commonly applied in the identification of Gram negative enteric bacilli, including members of the Salmonella, Citrobacter, and related groups. Each tube contains peptones, glucose, the amino acid lysine, and pH indicators such as bromocresol purple and cresol red. After inoculation, bacteria initially ferment the small amount of glucose present, producing acid that changes the medium from purple to yellow. If the organism possesses lysine decarboxylase, the enzyme converts lysine to cadaverine, an alkaline compound that raises the pH and returns the medium to purple. If the enzyme is absent, the medium remains yellow, indicating a negative reaction.

Nitrate Broth is a differential medium used to determine whether microorganisms can reduce nitrate to nitrite or further nitrogenous compounds through nitrate reductase activity. This biochemical test is commonly used for identification of Gram negative bacteria including Enterobacteriaceae, Pseudomonas, and certain Mycobacterium species.
